Isolation of New Transcription Factors that Interact with the Element (-38 to -22 bp) within the Minimal Promoter of ADH5/FDH Gene

Abstract
We have isolated 28 novel transcription factors which can bind to the element (-38 to -22 bp) located within the ADH5/FDH minimal promoter by screening a human spleen Gal4-activation domain fusion cDNA library utilizing yeast one hybrid system. The fusion proteins were able to activate transcription in vivo. Total cell extract prepared from the yeast transformed with Gal4-activation domain fusion cDNA library was able to bind to the element upon gel retardation assays. The size of the insert cDNAs ranged from 200 bp to 2 kb. Characterization by DNA sequencing and restriction mapping revealed that the clones 1, 4, 12, 20, 29, 17, and 8 are identical, but clones 20, 17 are slightly shorter in size. Most of the isolated clones are different from each other and code for new transcription factors. The clone 26 was highly homologous to the translationally controlled tumor protein, p21.
